摘要
目的 研究 型胶原、层粘连蛋白 (lam inin,L N)和间质金属蛋白酶 - 2 (matrix m etalloproteinase- 2 ,MMP- 2 )在不同年龄小鼠卵巢中的分布和变化模式。 方法 利用生物素 -抗生物素蛋白间接免疫荧光方法 ,从蛋白水平观察 型胶原和 L N的分布和变化 ;利用地高辛标记的 型胶原和 MMP- 2基因探针进行原位杂交 ,从m RNA水平观察 型胶原和 MMP- 2的分布和变化。 结果 出生 5 d小鼠卵巢的卵泡基膜和卵巢表面上皮(ovarian surface epithelium,OSE)基膜处已有少量 型胶原和 L N分布 ,基质细胞和 OSE中也有分布。 型胶原和 MMP- 2的 m RNA在基质细胞和 OSE有表达。发育期小鼠卵巢中卵泡基膜和 OSE基膜中 型胶原和 L N的含量随年龄的增加而呈增加趋势 ,分布也趋于连续。颗粒细胞、膜细胞、OSE中都有 型胶原和 L N分布 ,也有 型胶原和 MMP- 2 m RNA的表达 ,其表达水平明显比 5 d时高。成熟期小鼠卵巢 型胶原和 L N的含量较高。12个月的小鼠卵巢已开始退化 ,基膜细胞外间质 (extracellular matrix,ECM)的数量减少 ,MMP- 2 m RNA的表达量无明显变化。 结论 型胶原和 L N在小鼠卵巢发育过程中含量和分布发生着动态变化 。
Objective To investigate the distribution of type Ⅳ collagen and LN and the expression of type Ⅳ collagen and MMP 2 genes. Methods Biotin avidin DCS system indirect immunofluorescence technique, in situ hybridization with diglabeling type Ⅳ collagen and MMP 2 cDNA probe were used. Results In the ovary of 5 day old neonatal mouse, a small amount of type Ⅳ collagen and LN fluorescence was seen in the basement membrane of follicle and OSE. The expression of type Ⅳ collagen and MMP 2 was observed inside OSE and stromal cells. In 15day to 2 month old mouse ovaries, the contents of ECM components increased with advancing age, and the distribution of basement membrane componets were smoothly than before. The mRNA expressions of type Ⅳ collagen and MMP 2 was markedly increased during this peroid. OSE, granulosa and thecal cells were the cells, which contributed to the synthesis of those ECM components and MMP 2. In 2 to 6 month mouse ovaries, the amount of basement membrane ECM components did not change much, and maintain about the same level. In 12 month\|old mouse ovary, the contents of the basement membrane ECM components decreased. Conclusion We suggest that the type Ⅳ collagen and LN of ovary may play important roles in the process of follicle development.
