摘要
本工作在大鼠体内实验发现应用脂质体作为药物载体显著增强和延长脑钠素(BNP)的降压效应。体内代谢实验发现,应用脂质体作载体可以有效地延缓^(135)I-BNP在体内的降解。实验结果提示,应用脂质体保护BNP是提高BNP生物效应的有效措施之一。
Brain natriuretic peptide (BNP) has strong natriuretic, diuretic, vasodilative and hypotensiveaction, but in vivo it is easily degraded by peptidases. consequently its half life in blood circulationis very short, therefore its therapeutic usefulness is limited. This work found that injection into ratsBNP combined covalently with liposome, which incorporated peptidase inhibitors--thiorphan andcaptopril, the hypotensive effect of BNP was significantly intensified and prolonged, and that catabo-lism of ^(125)I-BNP in blood circulation was obviously delayed. These results suggest that the treatmentwith liposome as a drug-carrier is one of the effective method to elevate the biological effect ofBNP.
