摘要
本工作在大鼠体内实验,发现应用内肽酶抑制剂Thiorphan和/或二羧基肽酶抑制剂Capto-pril可以显著增强和延长脑钠素(BNP)的降压效应。体外实验发现Thiorphan和Captopril可以有效地抑制鼠肾组织提取液对^(125)I-BNP的降解。实验结果提示,应用酶抑制剂保护BNP是提高BNP生物学效应的有效措施之一。
Brain natriuretic peptide (BNP) has strong natriuretic, diuretic, vasodilative and hypotensiveaction, but in vivo it is easily disimilated, consequently its half life in blood circulation is short,therefore its clinical uses is limited. Thiorphan an endopeptidase inhibitor, and/or captopril a dicarbo-xylic peptidase inhibitor were injected together with BNP at the same time in rat, which significantlyintensified and prolonged the hypotensive action of BNP. When thiorphan or captopril was incubatedtogether with BNP in rat's kidney extract, it obviously inhibited the catabolism of ^(125)I-BNP. Theresults suggest that the usage of peptidase inhibitor to protect BNP in vivo is one of the effectivemethods to protentiate the biological effect of BNP.
