摘要
腺苷可使ROS 17/2.8细胞膜腺苷酸环化酶(AC)活性及膜流动性降低。腺苷与甲状旁腺素(PTH)合用,可显著地降低PTH对AC活性的激活程度,并同时使PTH降低碱性磷酸酶(AKP)活性的程度有所减弱。而当腺苷衍生物木糖腺苷或木糖腺苷8-六氢吡啶与PTH合用时却使AC活性大为增加,表明木糖基团的引入,可使腺苷对ROS 17/2.8细胞跨膜信息传递具有截然相反的作用。
The effects of adenosine and its analogs on signal transduction across plasma membrane in ROS17/2.8 cell line were observed. bPTH(1-34) increased the membrane fluidity and adenylate cyclase(AC) activity, which resulted in the increase of cAMP and the decrease of alkaline phosphatase(AKP) activity. Adenosine alone decreased the AC activity and membrane fluidity, these might bedue to the combination of adenosine with its inhibitory receptors at the plasma membrane, and/or bythe action of adenosine directly on the AC at the inner side of plasma membrane. But the combinationof adenosine and bPTH(1-34) can weaken the response of bPTH(1-34) on the AC and AKP activity.The opposite effects between adenosine and bPTH(1-34) may be beneficial to the maintenance ofnormal physiological conditions. Adenosine analogs-xylosyl adenosine (XylA) or xylosyl adenosine-8-hexylhydrogen-pyridine (XylA-P) had no effects on AC activity, but when combined with bPTH(1-34), both of them increased the AC activity markedly to 3-3.5 fold, far beyond the AC activitystimulated by bPTH(1-34) alone. It shows that xylosyl was the key group. Since cell proliferationcan be inhibited by intracellular level of cAMP, probably this is the machanism of antitumor ac-tion of XylA.
