克拉霉素对小鼠动脉粥样硬化炎症反应的影响研究

摘要 目的:探讨研究克拉霉素对小鼠动脉粥样硬化炎症反应的影响。方法:选择雄性的C57BL/6J小鼠40只为研究对象,通过建模分为对照组(10只)和实验组(30只),利用肺炎衣原体反复感染,同时给予高胆固醇食物饲养的方法,成功建立了动脉粥样硬化炎症模型。依据有无克拉霉素治疗及治疗的时间进一步分为未治疗亚组、早期治疗亚组、延迟治疗亚组各10只。数理统计分析各组的IL-6、IL-8水平及主动脉粥样硬化的治疗情况。结果:实验组小鼠的LDL-C、TG、TC水平显著高于对照组的(P<0.05),且均显示Cpn IgG抗体的阳性结果。与对照组相比,实验组小鼠的IL-6、IL-8水平显著升高(P<0.05)。实验组内IL-6、IL-8水平比较:早期治疗亚组、延迟治疗亚组的显著低于未治疗亚组(P<0.05),且早期治疗亚组的显著低于延迟治疗亚组(P<0.05)。未治疗亚组的动脉损伤率为100%,明显高于早期治疗亚组的20.00%及延迟治疗亚组的33.33%(P<0.05)。结论:早期克拉霉素治疗可以有效地减轻小鼠动脉粥样硬化炎症反应,延缓动脉粥样硬化的发生。 Objective: To study the impact of clarithromycin on atherosclerosis and inflammatory reaction in C57BL/6J mice.Methods: 40 healthy male C57BL/6J mice were divided into control group(n=10)and research group(n=30)through the modeling.And the research group was divided into three groups basis for whether clarithromycin treatment and treatment time: non-treatment subgroup(n=10),early treatment subgroup(n=10),and delayed treatment subgroup(n=10).The levels of IL-6,IL-8 and the condition of atherosclerosis were detected.Results: Atherosclerosis inflammation model was successfully established through chlamydia pneumomae(Cpn) repeated infection and high cholesterol food feeding.LDL-C,TG,TC levels of the research group were significantly higher than those in the control group with P0.05.And they are all display Cpn IgG antibody positive results compared with the control group.IL-6 or IL-8 level of the research group were significantly higher than those in the control group with P0.05.IL-6 or IL-8 level in the research group:non-treatment subgroup's was significantly higher than delay treatment subgroup' s;delay treatment subgroup' s was significantly higher than early treatment group's with P0.05.Arterial injury rate: non-treatment subgroup's was significantly higher than delay treatment subgroup's;delay treatment subgroup's was significantly higher than early treatment group's with P0.05.Conclusion: Clarithromycin has accurate clinical curative effect on atherosclerosis and inflammatory reaction in C57BL/6J mice.
