摘要
以解吸率为考察指标,通过单因素试验确定了AB-8型大孔吸附树脂纯化猴腿蹄盖蕨总黄酮的工艺条件.其最佳工艺条件为:猴腿蹄盖蕨总黄酮提取液的pH值为4~5、上柱液质量浓度为0.1g/mL、上样量为8.0 mg/g、吸附速率为1~2 BV/h、解吸液为75%乙醇、解吸速率为1~2 BV/h、解吸液用量为3 BV.在最佳纯化条件下,猴腿蹄盖蕨干浸膏中总黄酮的含量由19.21%提高到42.33%,说明上述纯化工艺条件在猴腿蹄盖蕨总黄酮的纯化方面具有一定的应用价值.
The purification conditions of total flavonoids in Athyrium Multidentatum (Doll.) Ching were determined through single factor experiments by using AB-8 macroporous absorption resin. The optimum conditions were as follows: the pH value of Athyrium Multidentatum extract 4 to 5, the mass concentration of loading solution 0.1 g/mL, the loading amount 8.0 mg/g, adsorption rate 1 to 2 BV/h, 75% ethanol as desorption solution, desorption rate 1 to 2 BV/h, and desorption solution amount 3 BV. Under the optimum conditions, the content of total flavonoids in dry A thyrium Multidentatum extract was improved from 19.21% to 42.33%. The results showed that the purification process had an application value in purifying total flavonoids in A thyrium Multidentatum (Doll.) Ching.
