摘要
目的观察丙酸睾酮注射液多次给药(28 d)对大鼠的毒性作用。方法雄性Wistar大鼠25只分为空白对照组、阴性对照组、丙酸睾酮注射液三个剂量组(6.125、12.5、25 mg)。阴性对照组给予相同容量茶油。一周给药3次,连续给药4周。观察大鼠一般状况、体重、血清生化及有关脏器系数等指标的变化,并对前列腺及注射部位进行病理学诊断。结果丙酸睾酮注射液给药组大鼠体重、血清生化与阴性对照组比较无明显差异;高、中、低剂量组胸腺脏器系数与阴性对照组比较有明显差异(P<0.01);高剂量组前列腺的脏器系数与阴性对照组比较有明显差异(P<0.01);中、低剂量组前列腺的脏器系数与阴性对照组比较有差异(P<0.05);前列腺镜检发现给药组部分大鼠的前列腺腺泡壁厚度略微增厚。结论丙酸睾酮注射液长期给药对大鼠的毒性较低,但可引起大鼠胸腺萎缩,前列腺增大。
Objective To observe the chronic toxicity of rats induced by Testosterone Propionate Injection. Methods 25 male rats were randomly divided into blank control ,negative control and three dose groups ,including 6. 125,12.5 and 25 mg per rat. The negative control group was treated with camellia oil at same amount. All the groups were intramuscularly ad- ministrated 3 times a week for 4 weeks. The changes of general behaviors, body mass increase, blood biochemistry, organs coefficient of rats were observed, and the pathological examination of prostate and injection sites were carried out. Results Compared with negative control group,the index of body mass and blood biochemistry of Testosterone Propionate Injection group had no significant difference and the organs coefficient of thymus had significant difference ,P 〈 0.01 ;The organs co- efficient of prostate of the high dosage group had significant difference, P 〈 O. 01, the low and medium groups also had differ- ence,P 〈 0.05. The pathological examination showed that the prostate wall in some drug - treated rat slightly thickened. Conclusion Long term administration of Testosterone Propionate Injection had little toxicity in rats but can cause atrophy of thymus and enlargement of the prostate.
