摘要
目的从花生中分离、纯化具有抗真菌活性的蛋白。方法提取花生粗提液,通过Affi-Gel Blue Gel亲和层析和SuperdexTM75分子筛层析,纯化得到目的蛋白。通过Tricine-SDS-PAGE、抗真菌活性检测鉴定该蛋白对不同真菌的抑制活性。结果对花生总蛋白进行SDS-PAGE电泳分析结果显示,其相对分子质量为12 000~70 000。组份C对绿色木霉有抑菌活性,对落花生褐斑病菌、黑曲霉、白地霉、产黄青霉均无抑菌活性。活性组份C层析得到的组份23对绿色木霉有抑菌活性。对活性组份23进行Tricine-SDS-PAGE电泳分析结果显示,组份23相对分子质量为24 000。结论花生中存在抗真菌活性蛋白。
Objective To isolate and purify the antimicrobial proteins from arachis hpyogaea.Methods Crude extracts were obtained from peanut and antimicrobial proteins were purified by affinity chromatography on Affi-Gel Blue Gel and gel chromatography on SuperdexTM 75 columns.The antifungal activity of the purified proteins was identified by Tricine-SDS-PAGE and antifungal activity test.Results SDS-PAGE showed that protein molecular mass ranged from 12 000 to 70 000.Component C could inhibit the growth of Trichoderma viride,but had no effects on Aspergillus niger,Geotrichum candidum,Penicillium chrysogenum and Mycosphaerella arachidicola.The ingredient 23,purified from component C,possessed inhibitory activity against Trichoderma viride.Tricine-SDS-PAGE showed that ingredient 23 had a relative molecular mass of 24 000.Conclusion Antimicrobial proteins isolated from arachis hpyogaea can prohibit the growth of some fungi.
