摘要
目的建立胶原诱导性关节炎(CIA)大鼠模型,并探讨胶原给药剂量以及性别因素对造模成功率的影响,优化造模方法。方法采用完全随机设计,以0、200、300、400μg 4个剂量水平的鸡胶原剂,免疫Wistar大鼠,连续观测大鼠一般情况、足爪容积、关节炎评分、血清抗CⅡ抗体、TNF-α浓度及大鼠关节病理情况。采用Logistic回归分析胶原蛋白剂量、性别与造模成功率的关系。结果发病大鼠其临床表现、免疫学及病理学特征显著,造模成功率随胶原蛋白剂量增加而上升(P<0.01,OR=14.414),性别对造模成功率无影响(P>0.05)。结论成功建立发病稳定的大鼠模型。在安全范围内,胶原蛋白给药剂量与造模成功率成正相关,性别对发病率无影响。
Objective To establish reproducible collagen-induced arthritis (CIA) models in rats, and optimize proce- dures for high incidence, in terms of the collagen doses and gender factor. Methods The Wistar rats were immunized by chick- en collagen type If (CII) to induce CIA. The rats in study were randomly divided into eight groups. Four different doses (400 μg, 300 μg, 200 μg, 0 μg) and gender factors were taken into account. Volume of paws was measured and arthritis severity scores were recorded. CII-specific antibody and TNF-a titers in serum were evaluated by ELISA. The pathological characteris- tics and bone erosion of the joints were checked by histopathologic technique. Logistic regression was used to analyze the rela- tionship of incidence and the two variates (doses of collagen and gender factors) Results CIA rats showed typical clinical, im- munological and histopathologic characteristics. The incidence went up with the dose of collagen (P〈0.01, OR: 14. 414), while gender factor showed no remarkable effect (P^0.05). Conclusion We have successfully established reproducible CIA models in rats with high incidence. Raising the dose of collagen can improve CIA incidence, while gender factor have no remark- able influence on it.
