摘要
以I-69杨试管苗和插条苗嫩叶为材料,直接用酶液处理不能获得高产率的原生质体。然而,将材料进行预处理,即可得到理想的原生质体分离效果。试管苗叶片经预处理后,产率每g鲜重可达3.05×10~7个原生质体;插条苗嫩叶经过预处理后,产率每g鲜重可达6.8×10~6个原生质体。预处理的作用可能主要是降低了叶片酚酸类物质的分泌。
It is difficult to get high-yield isolations of mesophyll protoplasts from the young leaves of shoot cultures and cutting shoots. In this investigation the leaves from shoot cultures and cutting shoots of P, deltoides Bartr. cv, 'Lux' (I-69-55) were both incubated with enzyme solution. Interestingly a high-yield isolation of mesophyll protoplasts can be obtained through pretreatment with an antibiotic. An everageof 3.05×10~7 protoplasts per g fresh weight were released from the leaves of shoot cultures,and 6.8 ×l0~B protoplasts per g fresh weight from the leaves of cutting shoots, The function of pretreatment may be to inhibit the leavessecretions of phenolic acids during incubation with enzyme solution.
