摘要
以SDS改进法、CTAB改进法及EASYAPIN试剂盒法3种方法提取山丹百合花蕾的总RNA,并通过紫外分光光度法和凝胶电泳检测对其提取效果进行分析比较。结果表明:SDS改进法提取的RNA效果不显著,CTAB改进法提取的RNA产量高,但有大量的DNA污染,需进一步纯化;EASYAPIN试剂盒法提取的RNA质量好,纯度高,完整性强,可直接进行下一步的分子生物学研究。
Three extraction methods（modified CTAB,modified SDS and EASYAPIN） were used to extract total RNA from flower bud of Lilium pumilum,and their effect were compared with UV spectrophotometry method and agarose gel electrophoresis detection.The results showed that no bands of RNA isolated by modified SDS method;the modified CTAB method could extract higher quality RNA from Lilium pumilum buds,but there was a large number of DNA contamination,without further purification;the total RNA extracted by EASYAPIN method could isolate high-quality and highly-complete RNA at a higher productivity and it was worth further biological research.
