摘要
目的:观察1-磷酸鞘氨醇(S1P)和溶血磷脂酸(LPA)对三维培养肺纤维细胞HFL-1凋亡的影响。方法:利用Ⅰ型鼠尾胶原制作含有HFL-1细胞的三维培养系统,分别把不同浓度的S1P(10-6~10-8mol/L)和LPA(10-5~10-7mol/L)加入培养液中,待胶原收缩5d后,测量胶的面积并加热使胶原溶解,采用TUNEL染色细胞核计算肺纤维细胞凋亡的百分数。结果:S1P和LPA剂量依赖性地引起含有肺纤维细胞的胶原显著收缩,并抑制细胞凋亡。结论:S1P和LPA有引起组织收缩和抑制其内的纤维细胞凋亡的作用,提示S1P和LPA可能参与肺组织重构,与肺纤维化形成有关。
Objective To observe the effects of sphingosine l-phosphate (S1P) and lysophospholipid acid (LPA) on the apoptosis of human fetal lung fibroblasts (HFL-1 ) in three dimensional collagen gels. Methods HFL- I s were cast into three dimensional culture system made with type I rat tail collagen, then different concentrations of SIP ( 10-6-10-8 mol/L) and LPA ( 10-5-107 mol/L) were added into the culture fluid. After contraction for 5 days, fibroblasts were harvested from the collagen gels by heating; then the apoptosis was assessed on cytospin preparations by TUNEL. Results Both S I P and LPA could stimulate the contraction of floating collagen gels, and could also inhibit the apoptosis of fibroblasts in 3D collagen gels in a concentration-dependent manner. Conclusions S1P and LPA may play a role in lung tissue remodeling and fibrosis formation by preventing fibroblasts apoptosis and the associated tissue contraction.
