摘要
为建立高效液相色谱法同时分离测定无糖乳制品中多种高倍甜味剂的含量,样品提取净化后,AtlantisT3C18色谱柱分离,甲醇:0.02mmol/L的硫酸铵溶液(v:v)=45:55等梯度洗脱,流速0.8mL/min,柱温30℃,二极管检测器205nm检测,进样量20μL。结果表明:糖精钠、安赛蜜、阿斯巴甜、甜蜜素4种甜味剂均能很好分离,糖精钠、安赛蜜、阿斯巴甜在浓度为1.00~20.0μg/mL,甜蜜素在浓度为0.05~0.5μg/mL范围内呈良好线性关系,RSD为1.7%~5.83%,平均加标回收率为85%~108%。该方法可同时对食品中的4种高倍甜味剂进行分离与检测,该方法灵敏度高,重复性好,为快速检测无糖乳制品中的高倍甜味剂提供技术支持。
A quantitive assay for the determination of four artificial sweetener in sugar-free dairy product was established by high performance liquid chromatography(HPLC).Sweetener was extracted and the separation was performed with Atlantis T3 C 18 Column,methanol:0.02 mmol/L(NH 4) 2 SO 4(v:v)=45:55 as mobile phase,flow rate:0.8 mL/min;column temperature:30 ℃.The whole separation process was completed at the UV wavelength of 205 nm by diode array detection,sample volume 20 μL.The result showed that the Acesulfame-K,Saccharin sodium and Aspartame showed distinct linearity relationship with the concentration in the range of 1~20 μg/mL,Sodium cyclamate showed distinct linearity relationship with the concentration in the range of 50~500 μg/mL,the correlation coefficient was 0.9942~0.9996,the recovery is 85.0%~108.0%,RSD was 1.7%~5.83%.In conclusion,the method was accurate,sensitive and feasible to analyze the content of artificial sweetener in sugar free dairy product.
