关节腔内注射CoCrMo纳米微粒对雄性大鼠生殖功能的影响

Effects of intraarticular injection of vitallium nanocomposites from metal-on-metal hip prosthesis on the reproductive function of male rats

摘要 目的探讨关仃腔内注射CoCrMo纳米微粒对雄性大鼠生殖功能的影响。方法利用直流电弧等离子体法制备CoCl·Mo纳米微粒。将75只8周龄雄性sD大鼠随机分为5组(每组15只):对照组膝关—肖腔内注射乍理盐水,基础量组、低剂量组、中剂量组及高剂量组分别注射剂量为50、250、500、1000μg/kg的CoCrMo纳米微粒生理盐水悬液,1次/周,连续10周。第11周处死动物,测定血清中co cr离子含量和睾酮水平,检测附睾精子的活力、精子计数、精子畸形率及精子凋亡率,观察睾丸组织的病理变化一结果基础量组、低剂量组、中剂量组及高剂量组血清co、cr离子含量呈上升趋势,均叫浪高于对照组;高剂堵组血清睾酮水平较对照组明显下降;中剂量组精子平均路径速度、鞭打频:举,以及乏高剂量绀精了平均路径速度、直线运动速度、曲线运动速度、鞭打频率明显低于对照组,以上各项目比较举异均有统计学意义(P〈0.05)。中剂量组精子计数、精子畸形率、凋亡率平均分别为(62.24±21.33)×l0/mL、12.1%±2.7%、12.4%±4.0%,高剂量组分别为(59.67±18.86)×106/mL、16.0%±3.8%、12.9%±4.4%,与对照组I(83.1)±23.58)x106/mL、6.2%±2.0%、6.1%±2.5%J比较蔗异均有统计学意义(P〈0.05)。基础量组、低剂量组睾丸组织无明显病理改变,中、高剂量组睾丸组织发生病理改变。结论父节腔内注射中、高剂量的CoCrMo纳米微粒可能对雄性大鼠生殖功能造成不良影响, Objective To inw;stigate the effects of intraartieular injection of vitallium nam^partieles from metal-on-metal hip prosthesis on the reproductive function of male rats. Methods CoCrMo nanopallicles were made by direct current arc plasma method. Most nanoparticles were round in shape and 50 nm in average size (range, from 20 to 100 nm). Seventy-five male Sprague-Dawley rats (8 weeks ohl) were randomly divi&.d into 5 even groups. In the control group, physiological saline was injected intraartic- ularly into the rats once weekly for 10 consecutive weeks; suspension with CoCrMo nanoparticles at different doses (50, 250, 500 and 1000 I.Lg/kg) was injected respectively in the basic, low, medium and high dose grot.ps in the same manner. At the end of the 1 lth week, the rats were sacrificed for measurement of serumal cobah aml chromium ion concentrations, testosterone level, epididymal sperm motility, and count, abnor- malily and apoptosis of sperms in all samples. Testicular tissues were harvested for pathological evahmtion. Results The serumal cobah amt chromium iml concentrations both increased in parallel with the increased doses of CoCrMo nanoparticles injected ( P 〈 O. 05). The serum testosterone level in the high dose group was significantly lower than in the control group; the sperm average path velocity (VAP) and beat cross frequency (BCF) i, the medium dose group as well as the VAP, straight line velocity (VSL), curvilinear velocity (VCI,) and BCF in the high dose group were significantly lower than in the control group ( P 〈 0. 05). The count, abncwmality and apoptosis rate of sperms were respectively (62.24 + 21.33) x 106/mL 12. 1% ±2.7% and 12.4%±4.0% in the medium dose group, and respectively (59. 67 ±18. 86) lOa/mL,16.0% ± 5.8% and 12. 9% ±4.4% in the high dose group, with significant differences from those in the control group [(83.11±23.58) 106/mL, 6.2% ±2.0% and6.1% ±2.5%] (P 〈0.05) . Obvious pathological changes in the testis tissues were observed in the medium and high dose groups but not in the basic and low dose groups. Conclusion Intraarticular injection of CoCrMo nanoparticles at medium and high doses may have negative effects on the reproductive function of male rats.
