不同强度电针对肥胖大鼠细胞因子信号转导抑制蛋白-3及过氧化物酶体增殖物激活受体-γ mRNA表达的影响 被引量:19

Effect of Different Intensities of Electroacupuncture Stimulation on Expression of SOCS-3 and PPAR-γ mRNA in Adipose Tissues of Obesity Rats

摘要 目的:观察不同电针强度对单纯性肥胖大鼠脂肪组织中细胞因子信号转导抑制蛋白-3(SOCS-3),以及过氧化物酶体增殖物激活受体-γ(PPAR-γ)mRNA表达的影响,探讨电针对细胞信号传导通路的调控机制。方法:SD大鼠随机分为普食组、模型组、5mA电针组、1mA电针组,每组10只,喂食高脂饲料造肥胖大鼠模型。电针组针刺双侧"足三里""三阴交",每天治疗1次,每次15min,共治疗2周。观察大鼠治疗前后体质量变化情况,反转录-聚合酶链反应法检测大鼠附睾脂肪组织SOCS-3及PPAR-γmRNA的表达。结果:造模后,各组大鼠体质量及SOCS-3、PPAR-γmRNA表达较普食组均明显升高(P<0.01);治疗后,两电针组体质量及SOCS-3、PPAR-γmRNA表达较模型组均降低(P<0.01),5mA电针较1mA电针效果更明显(P<0.01)。结论:电针"足三里""三阴交"穴对单纯性肥胖大鼠体质量及SOCS-3、PPAR-γ过度表达有良性调节作用,5mA电针比1mA电针效果更好。 Objective To observe the effect of electroacupuncture (EA) on the expression of suppressor of cytokine signaling 3 (SOCS-3) and peroxisome proliferator-activated receptor (PPAR)-γ genes in the epididymis adipose tissue of obesity rats so as to explore its mechanism underlying weight reduction. Methods SD rats were randomized into normal control, obesi- ty model, EA-5 mA and EA-1 mA groups (10 rats/group). EA stimulation (6 Hz/20 Hz, 5 mA or 1 mA) was delivered to the bi- lateral "Zusanli" (ST 36) and "Sanyinjiao" (SP 6) for 15 min, once everyday for two weeks. The expression of SOCS-3 mRNA and PPAR-γ mRNA in the epididymis adipose tissue was detected by reverse transcriptase polymerase chain reaction. Results Compared with the normal group, the body weight and expression of SOCS-3 and PPAR-γ mRNA in the model group were up-re- gulated obviously (P〈0.01). Compared with the model group, the body weight and expression of SOCS-3 and PPAR-γ mRNA were significantly decreased in the EA group, with the effects of EA-5 mA being better than those of EA-1 mA. Conclusion EA stimulation of ST 36-SP 6 can reduce body weight in rats with simple obesity, and the effect of higher intensity of EA is apparently better. Accordingly, EA intervention induces down-regulation of SOCS-3 and PPAR-γ mRNA expression in the epididymis adipose tissue.
