摘要
目的 了解阿萨希毛孢子菌(Trichosporon asahii,T.asahii)菌丝生长与顶体(spitzenkper)的关系。方法 用荧光染剂FM4-64对YPD液体培养基培养后的T.asahii进行荧光染色,经不同浓度细胞松弛素D抑制后,在荧光显微镜下观察染色结果。结果 在T.asahii芽胞、芽管以及菌丝顶端和亚顶端均可见到FM4-64荧光聚集;随着细胞松弛素D浓度的增加,其顶体荧光逐渐消失。T.asahii的生长受到明显抑制。结论 T.asahii存在与真菌极性生长密切相关的顶体,其在控制菌丝生长方面起着重要的作用。
Objective To understand the relationship between the growth of Trichosporon asahii （T.asahii） and spitzenkOrper. Methods Staining with the fluorescent dye FM4-64 and different density of inhibition of cytochalasin D on T.asahii, which cultured on YPD broth and staining results observed under a fluorescence microscope. Results Flourence of FM4-64 were found at the site of the apical vesicle cluster which named spitzenk6rper within spores, germ tubes and growing hyphal tips. The growth of Tasahii by the inhibition of cytochalasin D, and the higher the concentration of cytochalasin D, the less the hyphae of T.asahii.Conclusion Spitzenk6rper plays a vital role in controlling T.asahii polar growth.
