摘要
人胚脑组织标未经升主动脉灌注4%多聚甲醛和0.5%戊二醛。然后分别将其浸泡于上述固定液中3天和2周,将微量的DiI结晶植入视神经、额皮质和丘脑网状核内,室温下在同样固定液中贮存3个月和1年。经振动切片,磷缓贴片,置荧光显微镜落射光下观察。分别在视网膜额叶和背侧丘脑内均可见呈桔红色的荧光标记神经元。该技术为神经生物学研究开辟了前景。
A new fluorescent dye ,DiI was applied to the fixed human brain for
retrograde and an-terograde labeling of neuronal pathways. Four fetuses above 3 2 gestation
weeks collectedfrom legal abortion were perfused with 4%paraformaldehyde and
0.5%glutaraldehyde(in0.iM PB).The brain was removed from the skulls and post,fixed in the
saine fixative for 3days or 2 weeks,DiI crystals were placed into optic nerve,frontal coriex and
the thalamicreticular nucleus respectively.The tissue was stored in the above fixative in dark at
roomtemperature for 3 months or l year,then vibratomed. The sections were mounted with
0.1MPB· It is important to keep the sections from dryness. The sections were observed with
epi-fluorescent microscope equipped with a Rhodamine filter for viewing the orange-red DiI
fluo-rescence. There are many DiI labeling neurons in the retina,the frontal lobe and dorsal
thala-mus respectively
