不良孕产史夫妇男性精子染色体畸变情况研究 被引量:2

摘要 目的 研究男性精子染色体畸变状况以及生育情况。方法 采用三色荧光原位杂交(fluorescence in situ hybridization,FISH)方法,利用DNA染色体计数探针(chromesome enumeration DNA probes,CEP)X、Y、18与二硫苏糖醇(dithiothreitol,DTT)处理过的有不良孕产史夫妇(研究组)的男性(15例)精子核杂交,计数杂交信号,与对照组(生育过健康子女符合二胎生育指标想再生育的8名男性)进行比较。结果 研究组精子染色体X、Y、18二体率、X+Y缺体比率高于对照组(P<0.05),研究组精子的非整倍体染色体异常率高于对照组;研究组在避免接触有害物质和补充叶酸片剂等3~6个月后再行检查精子染色体X、Y、18的二体率和二倍体率,显示有所降低,差异有统计学意义(P<0.05)。结论 FISH技术是一种快速、准确、简单、实用性强的检测精子染色体畸变的方法。 Objective To research the relative factors of the male spermatozoon chromosome aberration in couples with unhealthy pregnancy and reproductive outcomes after intervene measures. Methods The technique of fluorescence in situ hybridization (FISH) for the chromosome enumeration DNA probes (CEP) of X, Y, 18 chromosome and Dithiothreitol(DDT) were used to hybridize the sperm nucleus of the unhealthy pregnancy couples and the hybridization signal was recorded and compared with control group. Results With chi - square test, the rate of sperm chromosome disome of X,Y,18 and nullisomic of X + Y of experimental group had exceeded the control group(P 〈 0.05 ). It was explained that was one of important reasons of unhealthy pregnancy for spermatozoon chromosome aneuploid aberration. By the intervene means of keeping harmful substance from themselves and taking orally folic acid for the male of the unhealthy pregnancy couples for 3 months -6months, it was significant reduced that the rate of sperm chromosome disome of X ,Y,18 and nullisomic of X + Y experimental group have been reduced. It was significant(P 〈0. 05). Conclusion The technique of FISH is the fast,accurate,simple,practical and useful method to detect the spermatozoon chromosome aneuploid aberration.
