亚甲基四氢叶酸还原酶C677T基因多态性与精子DNA碎片率的相关性研究 被引量:2

Correlation Between Methylenetetrahydrofolate Reductase C677T Gene Polymorphism and Sperm DNA Fragment Rate

摘要 目的研究亚甲基四氢叶酸还原酶(methylenetetrahydrofolate reductase,MTHFR)C677T基因多态性与男性患者精子DNA碎片率的相关性。方法观察1026例男性不育患者DNA碎片率,并检测其MTHFR C677T基因型。分析男性精子DNA异常发生率与MTHFR C677T基因型的相关性。结果不同基因型组间,DNA碎片率存在统计学差异(H=9.276,P=0.010),而在年龄差异无统计学意义(H=1.533,P=0.465)。精子DNA异常组和正常组的MTHFR C677T基因分布存在统计学差异(χ^(2)=9.897,P=0.007),男性突变纯合子基因型(TT型)与野生型(CC型)相比,发生精子DNA碎片异常的风险显著增加(OR=1.773,P=0.002)。结论MTHFR C677T基因型与男性精子异常相关,TT基因型是精子DNA异常发生的独立危险因素。 Objective To study the correlation between methylenetetrahydrofolate reductase (MTHFR) C677T gene polymorphism and male sperm DNA fragment rate in male patients.Methods Male sperm DNA fragmentation index and MTHFR C677T genotype of 1026 cases were observed.The correlation between abnormal incidence of sperm DNA and MTHFR C677T genotype was analyzed.Results There was significantly different in sperm DNA fragment index (H=9.276,P=0.010),while there was no significant difference in age (H=1.533,P=0.465) among different genotypes.There was significantly different in MTHFR C677T gene distribution between abnormal sperm DNA group and normal sperm group (χ^(2)=9.897,P=0.007).Compared with wild type (CC genotype),the risk of sperm DNA abnormalities of mutant homozygous genotype (TT genotype) was higher (OR=1.773,P=0.002).Conclusion These data suggest MTHFR C677T polymorphism is associated with male sperm abnormalities,and TT genotype is the independent risk for sperm DNA abnormality.
