养精种玉汤对胰岛素抵抗卵泡影响的实验研究 被引量:3

摘要 目的观察养精种玉汤复方水溶液对过多雄激素诱导的胰岛素抵抗(IR)卵泡胰岛素受体底物-1(IRS-1)和葡萄糖转运体-4(GLUT4)蛋白的影响,从而初步探讨其改善卵泡IR的机制。方法收集新鲜猪卵泡,经250μM丙酸睾酮(TP)作用48 h后,检测培养液上清残糖量,评估构建IR卵泡模型是否成功。采用蛋白印迹(Western Blot)检测IRS-1和GLUT4蛋白在正常组、抵抗组和复方组中的变化。结果 250μM的TP刺激48 h后,卵泡培养液上清残糖较正常组升高了29%(P<0.05)。当卵泡产生IR后,其IRS-1与GLUT4蛋白表达量较正常组分别降低了74%与95%(P<0.05)。然而加入养精种玉汤复方作用后,复方组IRS-1与GLUT4蛋白表达量较抵抗组分别升高了2倍与5倍(P<0.05)。结论过多雄激素可以使卵泡产生IR,而养精种玉汤能够上调产生胰岛素抵抗卵泡IRS-1与GLUT4蛋白的表达,这可能是其改善IR的机制。 Objective To observe the impacts of yangjing zhongyu compound formula solution on follicle insulin receptor substrate 1(IRS-1)and glucose transport body-4(GLUT 4)proteins in insulin resistance(IR)induced by excessive androgen so as to explore preliminarily the mechanism on the improvement in follicle IR.Methods The fresh follicles were collected from porcine ovary.After stimulated with 250 μM testosterone propionate(TP)for 48 h,the supernatant residual glucose content of the culture solution was detected.The success of IR follicle model structure was evaluated.Western Blot was adopted to detect the changes of IRS-1 and GLUT 4 in a normal group,an IR group and a compound group.Results After stimulated with 250 μM TP for 48 h,the supernatant residual glucose content of the follicle culture solution was increased by 29% as compared with the normal group(P〈0.05).After IR produced in the follicles,the expressions of IRS-1 and GLUT4 were reduced by 74% and 95% respectively as compared with the normal group(P〈0.05).After stimulated with yangjing zhongyu compound formula,the expressions of IRS-1 and GLUT4 were increased by 2 and 5 times respectively in the compound group as compared with the IR group(P〈0.05).Conclusion The excessive androgen may induce IR in the follicles.Yangjing zhongyu decoction up-regulates the expressions of IRS-1 and GLUT4 in IR follicles,which is probably the mechanism on IR improvement.
