摘要
目的:观察黄芩苷对apoE-/-小鼠的主动脉壁细胞内核因子κB(NF-κB)及ACE2蛋白表达的影响,探讨黄芩苷干预动脉粥样硬化(AS)进程的可能作用机制。方法:6周龄apoE-/-小鼠30只随机分成模型组和黄芩苷高低剂量组,同时用野生型小鼠C57BL10只作为正常对照组。正常组给予普通饲料喂养,模型组和黄芩苷组给予高脂饲料喂养12周,建立小鼠AS模型,黄芩苷组给予300 mg.d-1及150 mg.d-1灌胃8周;20周后处死小鼠。实验结束后,检测血脂,免疫组化分析主动脉壁细胞内NF-κB、血管紧张素转换酶2(ACE2)表达水平。结果:黄芩苷组与模型组相比,apoE-/-小鼠主动脉壁细胞NF-κB灰度值明显升高、ACE2灰度值降低有统计学差异(P<0.05)。结论:黄芩苷可能通过抑制NF-κB转录表达,干预炎症反应进而增强ACE2蛋白表达,干预动脉粥样硬化的发生发展。
OBJECTIVE To investigate the regulated effect of baicalin on NFkB and ACE2 protein expression in atheroscle rosis (AS) of ApoE-/- mice induced by hyperlipaemia. METHODS 30 healthy six weeks aged male apoE-/- rats were random ly divided into three group as follows: model group and the high and low does baicalin group, and 10 the wild type C57BL mice were used as the normal control group, as well. Normal group was given ordinary feed, the left three groups were given high fat feed on the basis of aortic balloon technique for twelve weeks to reproduce the rat AS model, and feeding with baicalin 300 mg.d-1 and 150 mg. d-1 eight weeks. After twenty weeks of experiment, blood lipids were detected, the NF-κB and ACE2 protein expression in aortic wall cells were measured by immunohistochemical methods. RESULTS The gray value of NF-κB protein expression in baicalin group was significantly reduced, however, ACE2 grey value was increased significantly, there were significantly difference compared with the model group. CONCLUSION Baicalin may enhance the stability of the plaque, and interpose the progress of atherosclerosis by inhibiting the NF-κB expression, elevating ACE2 expression.
