摘要
目的探讨细丝蛋白A(filamin A,FLNa)在胃癌组织中的表达水平及其临床意义。方法本研究应用逆转录聚合酶链反应(RT-PCR)和免疫组织化学方法(SP法),检测47例胃癌组织,及距胃癌组织边缘5 cm以上的显微镜下无癌浸润的正常组织中mRNA及蛋白的表达情况。结果 FLNa蛋白和mRNA在胃癌组织及正常胃组织中的表达阳性率分别为0.345±0.048、0.562±0.085和29.8%、78.7%,较正常胃组织明显降低(P<0.05)。结论胃癌组织中FLNamRNA和蛋白的表达较正常组织均降低,这可能与胃癌的发生、发展有关。因此,检测FLNa的表达可能对胃癌的临床治疗及预后判断有一定价值。
Objective To investigate the expression of filamin A (FLNa) in stomach carcinoma and its significance. Methods Reverse transcription polymerase chine reaction(RT-PCR) and Immunohistochemistry(SP) RT-PCR were respective-ly used to detect the expression of FLNa in 47 specimens of stomach carcinoma and normal stomach tissues ( 5 cm distant to the margin of lung cancer and no cancer infitration in microscope). Results The relative contents of FLNa mRNA were (0. 286 ± 0. 039 ), (0. 533 ± 0.087 ) in stomach carcinoma and normal tissues( P 〈 0.05 ). The positive rates of FLNa protein was 29.8% in stomach carcinoma and 78.7% expressed in normal tissues ( P 〈 0. 05 ). Conclusion The expression of FLNa mRNA and pro- tein are both lower in stomach carcinoma, suggesting that FLNa gene may be closely associated with oncogenesis, malignant de-gree. Detection of FLNa may be used for early diagnosis, clinical therapy and prognostic judgement of stomach carcinoma.
