Simultaneous Determination of Four Flavonoid Glycosides in Extracts of Folium Lithocarpi Polystachyi by HPLC

摘要 目的建立多穗柯提取物中根皮苷、3-羟基根皮苷、槲皮苷、山奈酚-3-O-α-L-鼠李糖苷的含量测定方法。方法采用HPLC法,Dikma PLATISIL TM-C18色谱柱(250×4.6 mm,5μm);流动相为甲醇(A)-0.2%磷酸水溶液(B),梯度洗脱(0~10 min,35%A→40%A;10~40 min,40%A→80%A),流速:0.8 mL.min-1;柱温:室温25℃;检测波长:260 nm。结果根皮苷、3-羟基根皮苷、槲皮苷、山奈酚-3-O-α-L-鼠李糖苷的线性范围分别为0.14~2.91μg(r=0.9998),0.82~16.48μg(r=0.9994),0.06~1.22μg(r=0.9999),0.05~0.98μg(r=0.9999);平均回收率分别为100.4%,99.5%,98.2%,97.3%(n=6)。结论该方法准确、简便,具有良好的重复性和稳定性,适合于多穗柯提取物中4种黄酮苷的含量测定。 Objective To develop a method for simultaneous determination of phloridzin, 3-hydroxyl phloridzin, quercetin, kaempferol-3-O-α-L-rhamnoside in the water extract of Folium Lithocarpi Polystachyi. Methods HPLC method was adopted. The analysis was performed on a Dikma PLATISIL TM-CI8 column (250 min×4.6 mm, 5 μm)with the mobile phase consisting of methanol(A)-0.2 % phosphate acid(B)by gradient elution(0-10 min, 35%A-- 40%A; 10-40 rain, 40%A→80%A)at a flow rate of 0.8 mL·min-1. The detection wavelength was set at 260 nm and the eolumn temperature was room temperature. Results The linear ranges were 0.14-2.91 ~zg(r=0.9998)for phloridzin, 0.82-16.48 μg (r=0.9994)for 3-hydroxyl phloridzin, 0.06~1.22 ~zg (r--0.9999)for quercetin, and 0.05- 0.98μg (r=0.9999) for kaempferol-3-O-α-L-rhamnoside. The average recoveries(n=6)of four components were 100.4 %, 99.5 %, 98.2 % and 97.3 %, respectively. Conclusion The developed method is rapid, accurate and with good repeatability. It is suitable for the quantitative analysis of phloridzin, 3-hydroxyl phloridzin, quercetin and kaempferol-3-O-cx-L-rhamnoside in the water extraet of Folium Lithocarpi Polystachyi.
