摘要
目的对钩吻根、茎、叶、花、果实生物碱进行薄层色谱研究。方法以钩吻碱甲和钩吻素子为对照,对不同产地钩吻的根、茎、叶、花、果实生物碱进行薄层色谱研究,考察了5种提取方法、4种展开系统、5种薄层板、3种检视方式以及不同温度和湿度对钩吻生物碱薄层色谱的影响。结果以方法 5提取,过PXC柱(阳离子交换树脂),以三氯甲烷-甲醇(9∶1)为展开系统,稀碘化铋钾试液-10%三氯化铁乙醇溶液(10∶1)混合溶液为显色剂,钩吻生物碱薄层色谱信息保留较多,分离较好,薄层效果最佳。结论该方法可作为鉴别钩吻的较专属鉴别方法。
Objective To establish a thin-layer chromatography(TLC)method for the determination of alkaloids in the root, stem, leaf, flower and fruit of Gelsemium elegans Benth. Methods With koumine and gelsemine as the refer- ence substances, 5 extraction methods, 4 developing systems, 5 TLC plates and 3 inspection modes as well as the factors of temperatures and humidity were studied in the assay. Results Method 5 was selected. After purified by PXC col- umn, the samples were developed by the solvent system of CHC13-MeOH (9 : 1), with thin potassium heptaiodobis- muthate test solution as color--developing agent. The method had much information of alkaloids, and had good resolution and retention. Conclusion The method can be used as a specific TLC identification method for Gelsemium elegans Benth.
