摘要
目的:利用人nm23-H1基因重组腺病毒转染Tca8113细胞,检测nm23-H1基因在Tca8113细胞中的表达。方法:制备人nm23-H1基因重组腺病毒并转染人舌癌Tca8113细胞系,采用Western blotting、免疫荧光法、免疫组化法和流式细胞术检测nm23-H1基因的表达。结果:Western blotting、免疫荧光法、免疫组化法和流式细胞术均检测到nm23-H1在Tca8113细胞中的表达,表达产物主要存在于细胞质中。结论:重组腺病毒载体Ad-nm23-H1成功介导了Tca8113细胞内nm23-H1基因的表达,为肿瘤的基因治疗提供了依据。
Objective: To analyze the expression of nm23-H1 gene in Tca8113 cells transfected by recombinant adenovirus. Methods: Tca8113 cells were transfected with Ad-nm23-H1. The expression of nm23-H1 gene in Tca8113 cells was determined by Western blotting, immunofluor-escence assay(IFA),immunohistochemistry assay(IHA) and flow cytometry. Results.. The expression of nm23-H1 protein was proved in Tca8113 cells by Western blotting, IFA, IHA and flow cytometry, and the expressed product mainly existed in cytoplasm. Conclusion:Ad -rim23-H1 successfully mediates the expression of nm23-H1 gene in Tca8113 cells which provided a basis for gene therapy of tumors.
