摘要 目的探讨GRP78及GRP94蛋白和mRNA在人胃癌组织中的表达及临床意义。方法收集胃癌手术标本183例,制成组织芯片,用免疫组化法检测GRP78和GRP94的蛋白表达;取30例胃癌新鲜组织用半定量逆转录聚合酶链反应(RT-PCR)检测GRP78、GRP94 mRNA相对表达。结果免疫组化法检测结果显示:GRP78、GRP94蛋白的阳性表达率分别为80.1%和73.8%,与癌旁正常胃黏膜比较差异有统计学意义(P<0.05);与肿瘤的大小、浸润、转移、分期有关(P<0.05);与性别、年龄和组织分化无关。半定量RT-PCR结果显示癌组织和癌旁组织中的GRP78、GRP94mRNA均有表达,胃癌组织的表达明显高于癌旁组织(P<0.05)。结论 GRP78、GRP94蛋白在人胃癌中呈高表达并与胃癌的转移和侵袭有关,可作为判断胃癌恶性程度的有用指标。GRP78、GRP94 mRNA的高表达可能与胃癌组织中的转录调控相关。 Objective To investigate the expression and clinical significance of protein and mRNA of GRP78 and G1RP94 in gastric carcinoma. Methods 183 samples were collected from the operation of gastric carcinoma and made into tissue microarrays. Then the protein expression of GRP78 and GRP94 was tested in immunohistochemical method. RT-PCR was used to test mRNA of GRP78 and GRP94 acquired from 30 surgical specimens of gastric carcinoma. Results According to the results of the immunohistochemistry, the positive rates of the protein from GRP78 and GRP94 were 80. 1% and 73.8% respectively, having statistical significance (P 〈 0. 05 ) with the adjacent normal gastric mucosa in comparative difference. They were related to the tumor size, depth of tumor invasion, lymph node metastasis and stage ( P 〈 0. 05 ), but irrelated to the gender, age and tissue differentiation. The results of semi-quantitative RT-PCR showed that there were expressions in mRNA from GRP78 and GRP94 in both cancer and adjacent tissues, and the expression of gastric carcinoma was apparently higher than that of adjacent tissues (P 〈 0.05). Conclusion The study suggests that the protein from GRP78 and GRP94,having high expression in the human gastric carcinoma, is related to gastric carcinoma metastasis and invasion and serves as a useful indicator of the diagnosis of gastric carcinoma. The high expression of mRNA from GRP78 and GRP94 is probably related to transcriptional regulation of gastric carcinoma tissues.
