摘要 目的:观察胃癌细胞株及人胃癌组织中Foxp3基因和蛋白的表达水平和分布特点,探讨胃癌细胞通过表达Foxp3直接参与免疫逃逸的机制。方法:应用实时荧光定量PCR技术,分别检测3株胃癌细胞株(MGC-803、BGC-823、SGC-7901)中的Foxp3mRNA表达水平,并采用流式细胞术破膜染色检测其Foxp3蛋白表达水平。对30例胃癌病理切片进行免疫组织化学染色,观察Foxp3在胃癌组织中的分布特点。结果:3株胃癌细胞株中均检测到有Foxp3基因表达,Foxp3的相对表达强度依次为BGC-823>SGC-7901>MGC-803,Foxp3蛋白表达水平与mRNA表达水平一致。30例胃癌病理切片中27例胃癌细胞表达Foxp3,且细胞质和细胞核都有表达。结论:Foxp3作为调节性T细胞发挥特异性免疫抑制功能的关键基因,在胃癌细胞中也有表达,这可能是其直接参与胃癌细胞免疫逃逸的重要机制之一。 Objective To investigate the expression of Foxp3 gene and protein in .gastric cancer (GC) cell lines and cancer tissue and explore the mechanism of GC cell immune escape mediated by Foxp3 expression. Methods Total RNA was extracted from three GC cell lines (MGC-803,BGC-823,SGC-7901) and the expression level of Foxp3 mRNA was determined by real-time PCR. The Foxp3 protein expression was determined by flow cytometry. GC tissue sections of 30 patients were stained by immunohistochemistry and the distribution patterns of Foxp3 in GC tissue were examined. Results Foxp3 mRNA was expressed in all the three GC cell lines, and the expression intensity was BGC-823〉SGC- 7901 〉MGC-803 respectively. The level of expression of Foxp3 protein correlated with the level of expression of Foxp3 mRNA. Expression of Foxp3 was observed in 27 of the 30 GC tissue sections, both in the nuclear and cytoplasm. Conclusions As a key gene of regulatory T cell mediated specific immune suppression, it is also expressed in GC cells. This may be one of the important mechanisms of immune escape of GC cell mediated by Foxp3.
