两种人脐静脉内皮细胞培养方法的比较 被引量:4

摘要 目的:用两种不同的方法培养人脐静脉内皮细胞,比较两种方法的优缺点。方法:(1)方法1,用0.125%胰酶加0.02%的EDTA消化液消化、方离脐静脉内皮细胞,用含20%的胎牛血清的DMEM完全培养液,于37℃、5%CO2条件下培养。(2)方法2,用胶原酶Ⅱ消化脐静脉内皮细胞,用含20%胎牛血清+细胞生长因子+明胶铺板,于37℃、5%CO2条件下培养。比较两种方法对细胞活力、培养细胞数量等方面的影响。结果:以改良方法培养的内皮细胞4h贴壁生长,细胞数量达到整瓶的80%~90%,48~72h生长最快,细胞融合,内皮细胞呈单层铺路石样外观。传统方法培养的内皮细胞24h后细胞数量只达到整瓶的20%~30%,细胞融合较慢。结论:用0.125%胰酶加0.02%的EDTA灌注脐静脉消化内皮细胞是获取内皮细胞的一种好方法,更加简便、经济,是建立稳定的人脐静脉内皮细胞分离和培养的好方法。
