摘要
目的:探讨TLR4、NF-κBp65、β-catenin、STAT3在溃疡性结肠炎癌变中的表达及作用。方法:60只SPF级Balb/c小鼠随机分3组:正常组、结肠炎组、结肠癌组,每组20只;除正常组外,模型组分别采用DSS和DSS/DMH复合法制备;应用显微镜观察结肠黏膜组织形态学改变,免疫组化法检测结肠黏膜TLR4、NF-κBp65、β-catenin、STAT3的表达。结果:结肠癌组小鼠结肠肉眼可见浆膜层散在、多处息肉状隆起变化,镜下见腺体萎缩、部分上皮内瘤变或癌变,其结肠组织TLR4、NF-κBp65、β-catenin、STAT3蛋白呈高表达特点;与正常组比较,差异有显著性意义(P<0.01)。结论:TLR4、NF-κBp65、β-catenin、STAT3蛋白及其信号通路在溃疡性结肠炎相关癌变发生发展中有重要作用。
Objective To explore the significances of the expressions of TLR4, NF-κB p65, β-catenin and STAT3 in the ulcerative colitis associated carcinogenesis (UCAC). Methods Sixty SPF Balb/c mice were randomly divided into three groups: normal group, UC group and UCAC group. Each group had 20 mice. The UC model was induced by oral administration of Dextran sulfate sodium (DSS), the UCAC model was induced by intraperitoneal injection of dimethylhydrazine (DMH) and oral administration of DSS. The colon histomorphology was observed by light microscope. The expressions of TLR4, NF-κB p65, β-catenin and STAT3 were detected by immunohistochemistry. Results The changes of hyperemia, edema and polyps occupation in colon placenta percreta of UCAC group were observed by naked eyes. Meanwhile, the changes of intraepithelial neoplasia and carcinogenesis were detected in UCAC group. The expressions of TLR4, NF-κB p65, β-catenin and STAT3 showed over-expression in UCAC group. Compared with normal group, the differences were obvious (P 0.01) . Conclusion The expressions of TLR4, NF-κB p65, β-catenin and STAT3 proteins and their signal transduction pathway may play important roles in the occurrence and development of UCAC.
