摘要
目的常春藤皂苷元体外对人结肠癌细胞LoVo增殖、粘附、侵袭和迁移能力的影响。方法采用MTT法、粘附实验、Transwell小室侵袭实验、划痕实验,观察常春藤皂苷元对人结肠癌细胞LoVo增殖、粘附、侵袭和迁移能力的影响。结果①常春藤皂苷元有抑制人结肠癌细胞LoVo增殖的作用,与对照组比较,随着药物浓度的增大,其对细胞的抑制率也明显增大(P<0.01),呈浓度依赖性。②常春藤皂苷元作用人结肠癌细胞LoVo后,细胞的粘附能力明显降低(P<0.01),随着浓度的增大和时间的推移,呈现抑制率递增的结果。③常春藤皂苷元作用人结肠癌细胞LoVo后,侵袭的细胞数较对照组明显减少(P<0.01),浓度越高,侵袭的细胞数量越少。④常春藤皂苷元作用人肠癌LoVo细胞后,细胞愈合的程度有明显的分组差别,浓度越高,愈合度越差,表明迁移能力受到很大影响。结论常春藤皂苷元对人结肠癌细胞LoVo的增殖具有抑制作用;常春藤皂苷元能抑制人结肠癌细胞LoVo的粘附能力、侵袭能力和迁移能力。
OBJECTIVE To explore the effect of hederagenin on the proliferation, adhesion, invasion and migration of hu- man colon cancer cell line LoVo in vitro. METHODS MTT assay, adhesion experiment, Transwell room invasion test, and e- rasion trace test were used to observe the effect of hederagenin on the proliferation, adhesion, invasion and migration. RE- SULTS (1)Hederagnin inhibited the proliferation of LoVo cells. Compared with control group, the inhibition ratio had been significantly increasing with the increase of the drug concentration, which showed a concentration dependence (P〈0.01). (2) Hederagnin remarkably reduced the adhesion capability of LoVo cells (P〈0.01), and along with the increasing drug concen- tration and time, the inhibition ratio increased. (3)Compared with control group, the number of invaded ceils was significantly decreased (P〈0. 01). The higher the drug concentration was, the less cells were invaded. (4)The cell healing degree had an obvious group difference, and the higher the drug concentration was, the worse the healing degree was, which indicated that migration ability was greatly affected. CONCLUSION Hederagenin can remarkably inhibit the proliferation, adhesion, invasion and migration of LoVo cells.
