摘要
目的探讨CIK细胞对人肺腺癌耐顺铂细胞A549/DDP体外杀伤效应及逆转作用。方法用MTT法分别检测顺铂、CIK细胞、CIK联合顺铂对人肺腺癌细胞A549及人肺腺癌耐顺铂细胞A549/DDP的杀伤活性,计算耐药倍数,逆转倍数。结果①顺铂对人肺腺癌细胞A549及A549/DDP的50%杀伤浓度(IC50)分别为1.823g/mL、6.392 g/mL,差异有统计学意义(P<0.05),人肺腺癌细胞A549/DDP对顺铂的耐药倍数(IR)为3.506。②CIK对人肺腺癌细胞A549及A549/DDP均有杀伤作用,其IC50分别7.657×105个细胞/mL、7.955×105个细胞/mL,差异无统计学意义(P>0.05)。③不同浓度的CIK细胞(1×105,2×105and3×105个/mL)与人肺腺癌细胞共培养后,再加顺铂,其IC50值分别降至1.743g/mL、1.336g/mL、0.883g/mL,差异有统计学意义(P<0.05);计算逆转倍数(FR)分别为(2.369、3.091、4.677)。结论①人肺腺癌细胞A549/DDP较A549对顺铂有明显的耐药性。②CIK对人肺腺癌细胞A549及A549/DDP均有杀伤作用,杀伤效果在两种细胞中无明显差异。③无毒剂量CIK联合顺铂作用于人肺腺癌细胞A549/DDP,可以增加顺铂对耐药细胞的杀伤活性。逆转倍数随着CIK密度的增加而增大,说明CIK细胞能部分逆转耐药细胞A549/DDP对顺铂的耐药性。
Objective To investigate the cytotoxic and resistance reversal effects of cytokine -induced killer cells (CIK) on cisplatin-resistant human lung adenocarcinoma cell line A549/DDP. Methods The cytotoxic ability of cisplatin, CIK cells and cisplatin combined with CIK cells was accessed by MTT assay followed by detecting the resistant index and the reversal fold. Results There was significant difference of the 50% inhibiting concentration (IC50) value between A549 and A549/DDP cells after cisplatin treatment. The resistant index of A549/DDP to cisplatin was 3. 506. Also, CIK had inhibitory effects on A549 and A549/DDP cells and no significant difference was shown between these 2 cells. Furthermore, after A549/DDP cells were pretreated with CIK at the density of 1 ×10^5 , 2×10^5 and 3 × 10^5 cells/mL and then treated with cisplatin, there were significant difference of IC50 value among these 3 different cell density of CIK cells with the reversal fold of 2. 369, 3. 091 and 4. 677, respectively. Conclusion ① A549/DDP cells are more resistant to cisplatin than A549 cells. ② There was no significant difference of the inhibitory effects of CIK cells on A549 and A549/DDP cells.③ CIK cells combined with cisplatin could increase the cytotoxic ability of cisplatin on A549/DDP cells, suggesting that CIK cells partly reversed the resistance of A549/DDP to cisplatin.
