摘要
评价对鲟源嗜水气单胞菌具有优良拮抗效果的解淀粉芽胞杆菌G1的安全性,以期推动其在水产养殖中的应用进程。通过特异PCR法检测了菌株G1的毒力基因,并采用纸片法分析了其对水产药物的耐药性,观察了其对小鼠和草鱼的毒力。结果表明,菌株G1不含有溶血素基因、肠毒素T基因、肠毒素FM基因;对诺氟沙星、恩诺沙星、新霉素、复方新诺明、甲氧嘧啶、链霉素、氧氟沙星、庆大霉素、复合磺胺等9种水产常用药物以及呋喃唑酮、环丙沙星、氯霉素、红霉素等4种水产禁用药物高度敏感;对四环素中度敏感。此外,菌株G1在105 cfu/g~109 cfu/g注射剂量下对小鼠和草鱼无致病性,对小鼠和草鱼的LD50大于109 cfu/g,为无毒菌株。
The present study was to evaluate the safety of t3. arnyloliquefaciens strain G1 against sturgeon-patho- genic Aeromonas hydrophila, and promote its application in aquaculture. The virulence genes of B. amylolique- faciens strain G1 were detected using specific PCR, its susceptibility against fishery and veterinary drugs was as- sayed using the disc diffusion method, and its pathogenicity to mice and grass carps was tested. The experimental results showed that the hemolysin (hlyA) gene and two enterotoxin (becT and entFM) genes were not present in the G1 strain. Strain G1 exhibited highly susceptible to 9 common fishery drugs (norfloxacin, enrofloxacin, neo- mycin, trimethoprim-sulfamethoxazole, sulfamethoxydiazine, streptomycin, ofloxacin, gentamidn, syntactic sul- fonamide) and 4 anti-doping fishery drugs (furazolidone, ciprofloxacin, chloramphenicol, erythromycin), interme- diately susceptible to tetracycline. In addition, the G1 strain was confirmed to be no pathogenic to mice and grass carps at the cell densities of 105 cfu/g-109 cfu/g body weight, and its lethal doses of 50% was estimated as above 109 cfu/g body weight. Thus, the G1 strain was safe in use.
