摘要
为探讨药源性胚泡不同生物学功能细胞损伤对孕 d8早期胚胎蛋白表达的影响 ,大鼠孕 d3分别 ig阿司匹林 (Asp) ,对乙酰氨基酚 (Par) 0 .2 5,0 .50 ,1 .0 g·kg-1,免疫外科术评价 Asp及 Par对孕 d4胚胎内细胞团 (ICM)细胞和滋养层细胞的发育毒性 ,十二烷基硫酸钠 -聚丙烯酰胺凝胶电泳评价Asp和 Par对孕 d 8胚胎蛋白表达的影响 .实验结果表明 Asp 0 .50 ,1 .0 g· kg-1组及 Par各剂量组ICM细胞数显著减少 (1 3.2± 2 .3,1 1 .1± 1 .7,及1 3.8± 1 .2 ,1 3.5± 1 .3,1 0 .0± 1 .4vs1 6.8± 1 .3,P<0 .0 1 ) .Asp1 .0 g· kg-1组及 Par各剂量均可诱导胚泡微核率增加 (P<0 .0 1 ) .Par呈剂量依赖性上调 d 8胚胎 70 ku蛋白表达的同时 ,下调小于1 4.4ku蛋白 (胚泡化特征蛋白 )表达 .提示 Asp及Par对大鼠 ICM呈选择性细胞毒作用 ,Par诱导孕d8胚胎 70 ku蛋白表达上调和胚泡特征化蛋白表达下调与 Par胚泡遗传毒作用相关 .
To explore the effects of the damages of two cell lineages with different biological functions from drug-exposed blastocysts on the protein expression of d 8 embryos, the developmental toxicity to inner cell mass(ICM) and trophectoderm(TE) on d 4 was evaluated after ICM was isolated by immunosurgery, and the expression of proteins on d 8 was identified after embryos were undergone sodium dodecyl sulfate-polyacrylamide gel electrophoresis after pregnant rats on d 3 of gestation were given ig aspirin(Asp) or paracetamol(Par) both at dose of 0.25, 0.50, and 1.0 g·kg -1. The results showed that the cell number of ICM in Asp 0.50 and 1.0 g·kg -1 groups and all the Par groups was markedly lower than that of the control (13.2±2.3, 11.1±1.7 in Asp groups, and 13.8±1.2, 13.5±1.3, 10.0±1.4 in Par groups vs 16.8±1.3 in the control, P<0.01). The frequency of micronuclei in blastocyst was increased both in Asp 1.0 g·kg -1 group(2.59%) and in all the Par groups(1.47%, 1.91%, 1.96%) vs 0.54% of the control(P<0.01). The increased expression of the protein with a molecular weight 70 ku concomitantly with the decreased expression of the proteins with molecular weight lower than 14.4 ku (blastocyst-specific proteins) was observed in Par groups dose-dependently. It is concluded that Asp and Par-induced cytotoxicity was more selective to ICM.
