摘要
初步研究了贵州习水国家级自然保护区土样放线菌。用改良HV培养基分离放线菌,对9株放线菌进行了形态学、生理生化和分子鉴定,系统发育分析和抗菌活性检测。结果显示:l株为小单孢菌属(Micromonospora),相似性97%;8株为链霉菌属(Streptomyces),相似性96%-99%。抗菌活性检测发现,链霉菌C2对3株供试细菌有抑菌作用,抑菌圈直径8.5~12mm:链霉菌C12对5株供试细菌具有抑菌效果,抑菌圈直径17~20mm。研究结果表明,小单孢菌为稀有放线菌,链霉菌为优势放线菌;C2和C12相比,C2抑菌作用弱,C12抑菌作用强,C12抑菌谱更广,效果更好。
To study actinomycetes of soils collected from Xishui National Nature Reserve in Guizhou, Actinomycetes were isolated from the soils by improved HV agar medium. Morphological, biochemical and molecular identification were proceeded on 9 selected actinomycetes. And phylogenetic analysis and antimicrobial activity test was done, too. The results showed that one was identified as Micromonospora (similarity 97%) and the other eight were identified as Streptomyces (similarity 96%N99%). Primary antimicromobial activity test showed that strain C2 was active against 3 test bacteria, the inhibition zone diameters varied from 8.5 to 12ram. Strain C12 was active against 5 test bacteria and the inhibition zone diameters vary from 17 to 20ram. Micromonospora was rare actinomycetes and Streptomyces was domain genera in the soils of Xishui National Nature Reserve. Strain C12 had more powerful and broader spectrum antibacterial activity than strainC2.
