摘要
目的 检验人参皂甙Rb1,Rg1和维生素E是否能抑制次黄嘌呤 (HX) /黄嘌呤氧化酶 (XO)对培养的牛视网膜色素上皮 (RPE)细胞的氧化损伤作用。方法 建立牛眼RPE细胞培养体系以及RPE细胞氧化损伤模型。利用MTT法观察人参皂甙Rb1、Rg1和维生素E对氧化损伤的牛RPE细胞增殖的影响。结果 加入不同浓度的Rb1后测得的细胞OD值普遍较HX/XO组的OD值高 ,但无显著性差异 ;当Rg1=0 1和 1μg/ml,维生素E =1和 10 μg/ml时测得的OD值与HX/XO组的OD值相比有显著性差异 (P <0 0 5 )。结论 Rg1和维生素E能对抗氧自由基对体外培养的牛RPE细胞生长的抑制作用 ,而Rb1的作用不明显。
ObjectiveTo investigate the effect of free radical damage on cultured bovine retinal pigment epithelial(RPE) cells,and identify anti free radical actions of ginsenoside Rb1,Rg1 and vitamin E.MethodsThe experimental model was based on the use of bovine RPE cells.RPE cells were isolated from bovine retina and maintained in monolayer culture.The cultured cells were confirmed by monoclonal antibody for anticytokeratin.Hypoxanthine/xanthine oxidase(HX/XO) and Rb1,Rg1 and vitamin E served as the radical generating system and scavengers,respectively.The effects of Rb1,Rg1 and vitamin E on oxidative damage of bovine RPE cells were determined by cell count and MTT method.ResultsThe culture cells had a typical morphology of RPE cells with a polygonal shape and light pigment.The RPE cells were evidenced by antibody cytokeratin and cytoplast of the cells show yellow granule stain.Exposure of bovine RPE cells to HX/XO inhibited proliferation at HX=0 1?mmol/L,XO=5?U/L.Application of Rg1(=0 1?&?1?μg/ml) and VitE(1?&?10?μg/ml) significantly increased the OD value in oxidized bovine RPE cells in comparison with HX/XO treatment ( P <0 05).No significant increasing was found between the OD values of Rb1 and HX/XO.ConclusionThe prevention of radical induced reduction of RPE cell growth by ginsenoside Rg?1 and vitamin E supports trials of therapy using antioxidents for various ocular syndromes with RPE involvement,as well as the use of Chinese traditional medicine in the treatment of these diseases.
