摘要
目的 比较角膜与三叉神经节 (TG)内单纯疱疹病毒Ⅰ型 (HSV 1)潜伏相关转录 (LAT)启动子序列 ,进一步证实角膜为HSV 1的另一潜伏地。方法 用新西兰白兔建立HSV 1潜伏感染动物模型 ,提取并PCR扩增角膜及TGLAT启动子 ,比较LAT启动子序列。结果 6 6 7% (4/6 )的角膜组织扩增出HSV 1LAT启动子 ,且与TG的电泳位置相同 (2 16bp) ;其在 2~ 16 1间的核苷酸序列完全相同。结论 HSV 1潜伏期的角膜组织存在LAT启动子 ,并与TG内的序列相同 ,表明角膜在很大程度上与TG一样 ,是HSV 1的另一潜伏点。
ObjectiveTo determine if cornea is another harboring latent of herpes simplex virus type 1(HSV 1),by sequences analysis of HSV 1 latency associated promoter in cornea and in trigeminal ganglion(TG).MethodsTotal DNA of cornea and TG from rabbits with quiescent herpes simplex keratitis (HSK) was extracted for PCR amplification followed by DNA sequencing.ResultsHSV 1 latency associated transcript(LAT)promoter was identified in 4 out of 6 corneas (66 7%),and its locations in ethidium bromide stained gels that from TG(216bp) DNA sequences between 2~ 161 nucleotides in cornea was identical to that in TG.ConclusionThe results suggest that the sequences of HSV 1 LAT promoter in cornea is identical to that in TG and both do not undergo rearrangement,giving further support to the concept of corneal latency.
