摘要
采用木霉为试验菌种、探讨了以麸皮为固态发酵基质产木聚糖酶及酶同步作用于麦麸以制备低聚木糖的可行性,为麸皮综合利用及低聚木糖制备提供新途径。考察了起始pH值、起始含水量、接种量、表面活性剂用量、发酵时间等因素对固态发酵的影响。通过正交试验确定最优发酵条件为:起始pH值3.0、起始含水量10mL、吐温-20添加量0.05%、木霉接种量1%。最优条件下发酵糖液可溶性总糖含量为16.492mg/mL,低聚木糖含量为11.088mg/mL,所得低聚木糖平均聚合度为3.052。通过薄层层析分析发酵糖液的主要成分为木三糖。综上,固态发酵麦麸制备低聚木糖方法具有可行性。
In this study, the preparation ofxylooligosaccharides (XOs) by Trichoderma sp. in solid-state fermentation using wheat bran as a sole substrate were investigated. The effects of initial pH, initial moisture content, inoculum, surfactant and fermentation time on fermentation were investigated. The optimal conditions were obtained by orthogonal experiment as follows: initial pH value 3.0, initial moisture content 10ml, Tween-20 0.05% and inoculum 1%. Under the optimum condition, the soluble total sugar content was 16.492mg/ml, the XOs content was 11.088mg/ml and the average de- gree of polymerization of XOs was 3.052. It was also found that main ingredient in fermentation broth was xylotriose detected by thin layer chro- matography. The results showed that. production of XOs from wheat bran by solid state fermentation was feasible.
