导向化疗对肿瘤细胞增殖动力学的影响 被引量:2

摘要 目的 :探讨导向化疗对肠癌细胞增殖动力学的影响。方法 :5 0例大肠腺癌患者随机分成三组 :A组单纯手术切除 ;B组术前静注化疗药物 ;C组术前静注导向化疗药物。术中取肿瘤组织及周围正常肠道组织进行AgNORs染色及检测。 结果 :1 手术组 (2 0例 )、化疗组 (13例 )、导向化疗组 (17例 )癌细胞AgNORs/核颗粒分别为4 18± 0 5 0、3 13± 0 2 0、2 2 9± 0 6 9,两两比较其差异存在显著性 (P <0 0 0 1) ;正常肠道细胞AgNORs/核均数分别为 1 81± 0 32、1 86± 0 2 5、1 80± 0 33,(P >0 0 5 )。 2 手术组、化疗组、导向化疗组肿瘤细胞AgNORs颗粒面积与细胞核面积的比值分别为 :0 2 85 8± 0 0 6 74、0 2 0 2 6± 0 0 5 93、0 2 0 0 9± 0 0 35 5 ,化疗组、导向化疗组与手术组比较其差异有显著性 (P <0 0 0 1) ,化疗组与导向化疗组之间的差异无显著性 (P >0 0 5 )。 3 手术组、化疗组、导向化疗组肿瘤细胞AgNORs颗粒光密度与肿瘤细胞核总光密度比值分别为 0 36 6 8± 0 0 6 0 3、0 30 36± 0 0 5 18、0 2 5 12± 0 0 374,两两比较差异有显著性 (P <0 0 1)。结论 :导向化疗与化疗均影响大肠癌细胞的增殖动力学 ,导向化疗对癌细胞增殖动力学的影响比化疗更明显。 Objective: To detect the effect of targeting chemotherapy on proliferous kinetics of colorectal carcinoma cells Methods:Fifty colorectal carcinoma patients were randomly divided into three groups:Group A,20 patients without preoperative chemotherap; Group B,13 patients receiving preoperative chemotherapy of 8 mg mitomycin C; Group C, 17 patients receiving preoperative targeting chemotherapy of 5 mg anti carcinoma embryo antigen monoclonal antibody C50 and 8 mg mitomycin C Resected specimens of colorectal carcinoma and adjacent normal tissue were assessed histologically by silver staining score of the nucleolar organizer region(AgNOR)to evaluate the effect to proliferous kinetics of tumor clonogenic cells Chemotherapy colorectal carcinoma with the values of vary kinds of AgNORs parameters,such as the average number of AgNOR per cell, the AgNOR/nucleoli area and the AgNOR/nucleoli luminous flux Results: The average number of AgNOR per tumor clonogenic cell in group A, group B, group C were 4 18±0 50, 3 13±0 20 and 2 29±0 69, respectively (P< 0 001) The average number of AgNOR in the normal colonic glandular cells in group A, B, C were 1 81±0 32, 1 86±0 25 and 1 80±0 33,respectively (P >0 05) The AgNOR/nucleoli area in group A, B, C was 0 2858±0 0674, 0 2026±0 0593 and 0 2009±0 0355, respectively, which was significantly different between group A and B, or group A and C (P< 0 001) but between group B and C (P >0 05) The AgNOR/nucleoli optical density(OD) in group A, B and C was 0 3668±0 0603, 0 3036±0 0518 and 0 2512±0 0374,respectively (P< 0 01) Conclusion: Both chemotherapy and targeting chemotherapy affect the proliferous kinetics of tumor clonogenic cells and reduce the proliferous speed The effect is stronger with targeting chemotherapy than with chemotherapy
