摘要
目的:检测Cyclin D1在腮腺多形性腺瘤及瘤旁涎腺组织中mRNA和蛋白水平的表达,探讨其在腮腺多形性腺瘤发生发展中的作用。方法:分别通过RT-PCR和Western blot方法检测Cyclin D1在原发腮腺多形性腺瘤与瘤旁涎腺组织中mRNA和蛋白水平的表达情况;另外采用免疫组化法检测Cyclin D1在原发腮腺多形性腺瘤以及瘤旁涎腺组织石蜡组织标本中蛋白水平的表达情况。结果:RT-PCR和Western blot检测结果显示,CyclinD1在组织标本中mRNA和蛋白水平不仅在于肿瘤组织中有较强的表达,而且瘤旁涎腺组织中Cyclin D1也有一定量的表达。免疫组化检测结果显示,Cyclin D1在腮腺多形性腺瘤组织中有较高的表达,而在瘤旁涎腺组织中未见明显表达。结论:Cyclin D1可能参与腮腺多形性腺瘤的发生发展,有助于为临床诊断治疗提供一定的参考依据。
Objective:To detect the mRNA and protein expression levels of Cyclin D1 in the parotid pleomorphic adenoma and adjacent tumor tissues,to explore the role of Cyclin D1 gene in pleomorphic adenoma.Methods:RT-PCR and Western Blot were used to detect the mRNA and protein expression levels of Cyclin D1 in primary parotid pleomorphic adenoma and adjacent tumor tissues.Immunohistochemistry was also applied to detect the expression of Cyclin D1 in paraffin embedded primary parotid pleomorphic adenoma tissues and adjacent tumor tissues.Results:As shown by RT-PCR and Western blot,Cyclin D1 expressed both in tumor tissues and adjacent tumor tissues of pleomorphic adenoma.Compared to its higher expression in tumor samples,the expression of Cyclin D1 in adjacent tumor tissues was relatively weaker.However,immunohistochemical results showed that Cyclin D1 was restrictively detected in tumor tissues of parotid pleomorphic adenoma,but none in adjacent tumor tissues.Conclusion:Cyclin D1 may play a role in the occurrence of pleomorphic adenoma.The detection of Cyclin D1 might provide information for prognosis and treatment of parotid pleomorphic adenoma.
