脂联素启动子单核苷酸多态性频率及其水平与2型糖尿病的相关性 被引量:4

The study on the association of adiponectin promoter single nucleotide polymorphism frequencies and adiponectin levels with type 2 diabetes mellitus

摘要 目的探讨血清脂联素(APN)水平及APN基因启动子区域单核苷酸多态性(SNP)频率与2型糖尿病(T2DM)之间的相关性,观察脂联素基因(aPM1)启动子多态性是否为T2DM的遗传危险因素。方法对单纯T2DM组(A组)、T2DM合并肾损害组(B组)及正常对照组(C组)分别测定血清APN水平及生化指标。采用ELISA法检测血清APN水平,并与其他生化指标进行相关性分析。应用变性高效液相色谱(DHPLC)技术筛选aPM1多态性位点。结果 C组APN水平明显高于A、B组(P<0.01);血清APN水平与高密度脂蛋白(HDL)呈正相关(P<0.01),与空腹血糖(FBG)呈负相关(P<0.01);与胆固醇(CHO)、甘油三酯(TG)等均无显著相关性。在aPM1启动子区-11377位点存在SNP(GG、GC、CC),G等位基因携带者患T2DM的风险率显著高于C等位基因携带者(OR=2.09,P=0.028)。结论 APN可能具有改善胰岛素抵抗的作用,并且可能是心血管的保护因子;aPM1-11377C/G多态性与T2DM发生、发展有相关性,提示SNP-11377C/G多态性可能增加T2DM的遗传风险。 Objective To investigate the association of the levels of serum adiponectin(APN) and the APN gene(aPM1) promoter region single nucleotide polymorphism(SNP) frequencies with the type 2 diabetes mellitus(T2DM).And it is expected to discover whether the aPM1 promoter polymorphism is a genetic risk factor for T2DM.Methods Serum APN and biochemistry items were measured in the simple T2DM group,the T2DM with renal impairment group and the healthy control group.The serum APN levels were determined by ELISA.The correlation between the serum APN levels and other biochemical parameters was analyzed.And the aPM1 polymorphisms were screened by the technique of denaturing high performance liquid chromatography(DHPLC).Results The serum APN levels of the healthy control group were significantly higher than that of the simple T2DM and the T2DM with renal impairment group(P0.01).And the serum APN levels were positively correlated with the high density lipoprotein(P0.01);negatively correlated with the fasting plasma glucose(P0.01);there was no significant correlation with cholesterol and triglyceride.SNP(GG,GC,CC) was presented at the promoter region of the aPM1-11377 sites.And the risk of suffering of T2DM in G allele carriers was significantly higher than the C allele carriers(OR=2.09,P=0.028).Conclusion The APN may improve the insulin resistance.It may be the cardiovascular protection factor.And aPM1-11377C/G polymorphism is associated with the occurrence and the development of T2DM.It is indicated that the SNP-11377C/G polymorphism may increase the genetic risk of T2DM.
