cAMP对人骨髓瘤细胞的IL-6信号转导功能及细胞增殖的抑制作用 被引量:3

摘要 目的 研究PKA途径激活 (或胞内cAMP水平升高 )后对人骨髓瘤细胞的IL 6信号转导功能及细胞生长的影响。方法 采用PKA途径激动剂Foskolin(FK)和cAMP类似物 8 Br cAMP刺激人骨髓瘤细胞系———Sko 0 0 7,分别通过MTT方法和凝胶阻滞电泳 (electrophoreticmobilityshiftassay,EMSA)方法检测FK和 8 Br cAMP对Sko细胞的生长及其IL 6信号转导功能的影响。结果 FK和 8 Br cAMP可对Sko细胞的生长及其IL 6的Jak STAT信号途径的活化起双重抑制作用 ,但对另一条IL 6信号途径———Ras NF IL 6途径的活化无显著影响。结论  1.PKA途径的激活可调控骨髓瘤细胞系中IL 6的Jak STAT信号转导途径 ;2 .Sko细胞中PKA途径激活所介导的细胞增殖抑制作用与IL 6的Jak STAT信号途径的活化抑制作用相关。 Objective To investigate the effect of PKA pathway activation (or increasing level of intracellular concentration of cAMP) on the growth and IL 6 signal transduction of the multiple myeloma (MM) cells. Methods Human multiple myeloma (MM) cell line Sko 007 were stimulated with the angonist of PKA forskolin(FK) or the analogue of cAMP 8 Br cAMP, the effect of FK and 8 Br cAMP on cell proliferation and IL 6 signal transduction of Sko 007 cells were exhibited by MTT and electrophoretic mobility shift assay(EMSA). Results FK and 8 Br cAMP inhibited the proliferation and Jak/STAT pathway activation of Sko 007 cells, but had no regulatory effect on Ras/NF IL 6 pathway. Conclusion 1.Jak/STAT pathway activation by IL 6 in MM cell line Sko 007 can be regulated by cAMP. 2.The inhibitory effect on the proliferation of Sko cells mediated by the activation of PKA pathway is temporally related to the inhibition of JAK/STAT pathway activation by IL 6.
