摘要
在碱性条件下,4-硝基重氮苯与盐酸异丙肾上腺素发生偶合反应,生成一种黄绿色产物。基于这一原理,建立了顺序注射分光光度法快速测定盐酸异丙肾上腺素的新方法。通过计算机控制系统程序,数据的采集和处理用FIAlab5.0软件进行。系统研究了顺序注射控制程序、进样顺序、反应管及流速、多种试剂的注入体积和浓度的影响以及pH值的影响。在最佳条件下,方法的线性范围是0.050~1.0μg/mL,检出限为0.03μg/mL,每小时可进样36次,每次分析仅消耗试剂0.105mL。相对标准偏差为3.0%(0.8μg/mL盐酸异丙肾上腺素,n=11)。干扰实验发现,一些常见的化学物质和药物添加剂不影响测定,本方法已用于水样、尿样、药物中异丙肾上腺素的测定,回收率在96.3%~103%之间。t-检测证明与药物标示量相符。
In basic medium, isoprenaline hydrochloride responsed with nitroaniline to product yellow green azocompound. Based on this mechanism, a new method of sequential injection spectrophotometric for rapidly determination of isoprenaline hydrochloride was established. Through the program design, FIAlab5.0 software was used for data collection and processing. The influence of parameters were systematically investigated including the influence of sequential injection control program, the aspiration sequence, the reaction coil and flow rate, dosage of reagent and the pH value. Under the optimum experimental conditions, the linear range of the calibration curve for isoprenaline hydrochloride was 0.050-1.0 μg/mL and the detection limit of 0.03 μg/mL was achieved. The sample throughput was 36 h-1. The total reagent consumption in a procedure was only 0.105 mL. The relative standard deviation was 3.0% (0.8 μg/mL isoproterenol hydrochloride was used for the determination, n = 11). In the interference experiments, the determinations were not affected by common ions and pharmaceutical excipients. This method was used for the determination of isoprenalinehydrochloride of water,urine and drugs with the recoveries between 96.3 %-- 103 %. Statistical analysis revealed that there was no significant different at drug labeled amount.
