摘要
目的探讨复乳法制备丝裂霉素C(MMC)脂质体的工艺方法和包封率测定。方法采用复乳法制备MMC脂质体,用透射电镜观察所制备的MMC脂质体的形态,选择MMC的吸收波长,葡聚糖凝胶柱分离游离MMC和MMC脂质体,测定脂质体混悬液中的总药物量与游离药物量,完成MMC脂质体包封率的测定。结果所制得的MMC脂质体呈球形与类球形,未过柱除去游离药物的MMC脂质体混悬液中MMC的浓度为341.16μg/mL,用柱层析分离除去游离药物后所得的MMC脂质体定容至一定体积,其中药物浓度为1.41μg/mL,MMC脂质体平均包封率为8.25%。结论复乳法制得的脂质体的包封率较高,而且方法简单易行,药物产率也较高。
Objective To study a method for determination of entrapment efficiency of liposomal Mitomycin C(MMC)by double emulsion.Methods Double emulsion was used to prepare liposomal MMC.The shape of liposomal MMC was observed under transmission electron microscope and the wavelength of MMC was chosen.MMC and liposomal MMC were separated by Glucose gelcolumn chromatography and the entrapment efficiency of liposomal MMC were determined.Results The shape of liposomal MMC was spherical.The concentration of MMC in liposomal MMC suspension which with free medicine was 341.16 μg/mL.After separating free medicine by Glucose gelcolumn chromatography,the concentration of liposomal MMC was 1.41 μg/mL and the entrapment efficiency of liposomal MMC was 8.25%.Conclusion The entrapment efficiency of liposomal MMC by double emulsion is high.The method is simple and the productivity of medicine is high,too.
