摘要
用猪睾丸细胞(ST细胞)从1只发病仔猪的脑组织中分离到1株病毒,第6代时的毒价为10^(7.75)TCIDs_(50)/mL。用猪伪狂犬病病毒特异性引物可从该病毒细胞培养物中扩增出与预期大小相同的gB基因部分核苷酸片段。测序结果表明:该gB基因的部分序列与GeneBank登录的5株PRV同源性为98.0%~99.5%。接种家兔后可观察到典型的伪狂犬病临床症状,ELD_(50)为10^(-7.50)/mL,分离毒可被PRV阳性血清中和。该分离毒为猪伪狂犬病病毒强毒株。
A virulent strain of pseudorabies virus (PRV)from the brain tissue of a diseased piglet was isolated with swine testis cells(ST cells),and its virus titer at the 6th passage was 107.75 TCID50/mL. By using specific primers of porcine pseudorabies virus,a partial nucleotide fragment of gB gene the same size as expected was amplified from the virus cell culture. The sequencing showed that the homology between the partial sequence of gB gene and those of 5 strains of PRV registered in the GenBank was 98.0%- 99.5%. The typical clinical symptoms of pseudorabies could be observed in the inoculated rabbits and the ELD50 was 10-7.50/mL. The isolated virus could be neutralized by the positive serum of PRV.
