摘要
为了探讨双酚A(BPA)内分泌干扰效应,研究了BPA对斑马鱼肝脏雌激素受体(Esr1 mRNA)、芳基烃水解酶受体(AhR2 mRNA)以及卵黄蛋白原(VTG1 mRNA)表达的影响。结果表明:BPA暴露12 h后,Esr1 mRNA和VTG1显著提高(P<0.05)。BPA暴露24 h后,Esr1 mRNA和VTG1 mRNA相对表达量依1日保持相当高的水平(P<0.05)。暴露48 h后,Esr1 mRNA的相对表达量继续升高。暴露72 h后,Esr1 mRNA和VTG1 mRNA相对表达量低于暴露12、24 h和48 h。暴露48 h后,AhR2相对表达量为0 h的3.22倍。暴露过程中,Esr1 mRNA和VTG1 mRNA相对表达量变化一致。BPA对雄性斑马鱼的Esr1 mRNA和VTG1mRNA诱导能力较强,但对AhR2诱导能力较弱。表明BPA对斑马鱼的内分泌干扰作用存在多途径过程。
In order to better understand the endocrine disrupting effect of bisphenol A(BPA)on ze- brafish,the expressions of Esr1 mRNA(ERα),AhR2 and VTG1 mRNA in male zebrafish livers were studied in being exposed to BPA. The results showed that 12 h later,the Esr1 mRNA and VTG1 signif- icantly increased(P〈0.05);24 h later, the relative expression levels of both Esr1 mRNA and VTG1 mRNA remained quite high(P〈0.05) ;48 h later,the relative expression level of Esr1 mRNA contin- ued to rise;the relative expression levels of both Esr1 mRNA and VTG1 mRNA at 72 h were lower than those at 12,24 and 48 h. During being exposed to BPA for 96 h,the relative expression level of AhR2 mRNA 48 h later was 3.22 times as high as that at 0 h,and both Esr1 mRNA and VTG1 mRNA relative expression levels were not different in variation. The inductive capacity of BPA for the Esr1 mRNA and VTG1 mRNA was greater but that for the AhR2 was less. It was concluded that the endo- crine disrupting effect of BPA on zebrafish could be a process of multiple pathways.
