摘要
对杭白菊中的抗胃癌活性成分进行了半仿生提取,获得了具有抑制胃癌细胞生长的有效组分及最佳提取工艺.采用单因素实验考察了提取时间、料液比、胃蛋白酶用量及胰蛋白酶用量等因素对杭白菊提取物抑制胃癌细胞生长活性的影响,并采用均匀设计法系统优化了杭白菊抗胃癌活性成分的半仿生提取工艺.实验结果表明:杭白菊半仿生提取物确实具有抑制胃癌细胞生长的作用.最佳提取工艺:提取时间1h,料液比1∶12,胃蛋白酶用量为0%,胰蛋白酶用量为0%.此工艺获得的杭白菊提取物处理下,胃癌细胞存活率为19.9%.
In this paper, semi-bionic extraction method was used to extract the components against stomach cancer from chrysanthemum, and the active components and the optimum extraction conditions were achieved. The influences of extraction time, ratio of material to extraction liqiud, and concentrations of pepsin and tyrosine on the activity of chrysanthemum extracts against stomach cancer cells were investigated by single factor experiment, and the extraction process was optimized by uniform experimental design method. The results showed that chrysanthemum extracts exerted inhibiting activity on the growth of stomach cancer cells, and the viability of stomach cancer cells treated with chrysanthemum extracts was 19.9%. The optimum extraction conditions: extraction time 1 h, ratio of mat the concentration of pepsin O~ and the concentration of tyrosine erial to extraction liquid 1 : 12, 0%.
