摘要
将囊胚细胞核直接移植到未去核的斑马鱼未受精卵子中 ,首次获得了正常发育的幼鱼。染色体检查结果显示移核胚胎众数为二倍体 ;组织切片检查结果表明卵子中未去掉的原有的雌原核在供体核移入之后退化 ,不参与胚胎发育。细胞核移植在模型鱼斑马鱼上迄今未见报道 ,核移植方法不但简单省时 ,而且供体胚胎仍可正常发育 ,为移核胚胎提供理想的对照。
WT5BZ]Nuclei from zebrafish blastulae were transplanted into the non enucleated eggs of the same species. The recipient eggs cleaved normally, and could give rise to larval fish. The chromosome number analysis showed that the majority of the recipient embryos were diploid. The histological examination also revealed that the original female pronuclei in the recipient eggs became degenerated at early cleavage stage. It suggested that they did not take part in embryogenesis. This is the first report on cloning of zebrafish. The nuclear transplantation methodology described in this study is not only simple but also time saving. In addition, the donor embryos are maintained to develop normally, and thus provided an ideal control for the development of recipient embryos. Cloning or nuclear transplantation of zebrafish (Brachydanio rerio) within same species has not been reported thus far.
