白芍总苷对正常人和SLE患者PBMC分泌IFN-α的影响 被引量:3

Influence of Total Glucosides of Paeony on Level of Interferon-alpha in Healthy Donors and Patients with Systemic Lupus Erythematosus

摘要 目的了解白芍总苷(TGP)对正常人和系统性红斑狼疮(SLE)患者和外周血单个核细胞(PBMC)分泌干扰素(IFN)-α的影响,探讨TGP治疗SLE的作用机制,为TGP治疗SLE提供理论依据。方法选取正常人30例(正常对照组)和SLE患者60例(SLE组),抽取外周静脉血约10 ml,分离外周血单个核细胞(PBMC),再分为2组体外培养。正常对照组分为:a组,为空白对照组;b组,加入TGP。SLE组分为:A组,为空白对照组;B组加入TGP。各组均培养48 h后提取细胞培养上清液,用ELISA方法检测PBMC培养上清中IFN-α的浓度,分别比较正常对照组与SLE组PBMC加入TGP培养与空白对照组之间培养上清液中IFN-α的分泌水平有无差异。结果体外培养后细胞上清液IFN-α的水平变化如下:a组为(1.42±0.21)μg/ml,b组为(1.14±0.17)μg/ml,b组显著低于a组(P=0.048);②A组(4.83±0.53)μg/ml,B组(3.23±0.45)μg/ml,B组显著低于A组(P=0.001)。结论 TGP可以抑制SLE患者和正常人PBMC分泌IFN-α,治疗SLE可能有效。 Objective To investigate the influence of total glucosides of paeony (TGP) on the expression of the secretion of interferon-alpha(IFN-α) of peripheral blood mononuclear cells (PBMC) in patiengs with systemic lupus eiythematosus(SLE) and normal donors. Methods Thirty healthy donors (as control) and sixty SLE patients (study group) were enrolled in this stud- y, and the peripheral blood mononuclear cell(PBMC) was isolated, PBMC were divided into 2 groups. In the control group, group a was free of TGP, group b incubated with TGP. According to the above method, study group were divided into group A and group B. Each group were cultured respectively for 48 hours. ELISA was used to detect the secretion of IFN-o~ in supematants. SPSS 13.0 software was used to statistical analysis. Results The concentration of IFN-αin the supernatants : (1)group a was ( 1.42 ± 0.21)l.Lg/ml,group b was(1.14 -±0.17) p.g/ml;the concentration of IFN-α in group b was lower than that in group a(P 〈 0.05 ). (2)group A was ( 4.83 ± 0.53 ) p,g/ml, group B was ( 3.23 ± 0.45 ) μg/ml ; the concentration of IFN-α in the group B was lower than that in group A ( P 〈 0.01 ). Conclusion TGP can inhibit secretion of IFN-α of PBMC of healthy donors and the pa- tients with SLE and it may be effective to treat SLE.
