摘要
目的 通过测定分析烟碱诱导出现的差异表达片段的序列,观察与烟碱相关的动脉粥样硬化的致病基因。方法 提取以烟碱处理培养的胎儿脐静脉内皮细胞的RNA,利用mRNA差异显示技术显示差异表达cDNA片段(EST),经Northern 印迹证明并测序,经基因库比较查询。结果 发现60条EST,Northern 印迹证实23条cDNA确属差异表达片段,测序发现11条未见报道同源序列的EST。结论 确定了11条差异表达片段的序列,为全长cDNA筛选和克隆打下基础。
Objective To identify the sequences of cDNA fragments that were differentially expressed between normal human umbilical vein endothelial cells (HUVEC) and HUVEC stimulated by nicotine. Methods The differential display reverse transcription polymerase chain reaction was used to search for differences between the mRNA expression of HUVEC and HUVEC induced by nicotine. Some differentially expressed cDNA fragments (ESTs) were found, which was further comfirmed by Northern blotting, and then compared with Gene bank. Results Sixty cDNA differentially expressed fragments (ESTs) were displayed, among them, 23 were reconfirmed by Northern blotting. In comparison with Gene bank, 11 novel ESTs were obtained. The article reported the results of one of them, CY31. Conclusion Eleven novel ESTs were ascertained, which provides a basis for our further research.
