摘要
本文介绍一种测定各种饲料蛋白质含量的新方法—蛋白质水解茚三酮法。将标准蛋白和几种饲料样品加3%H_2SO_4,直接加热6小时,通过蛋白质水解产生的氨基酸与茚三酮反应的光吸收值,计算总蛋白量;与未水解的标样及饲料样品的甲醇提取液的茚三酮反应吸收值之差计算去游离氨基酸的纯蛋白质量。本法与凯氏氮素定量法无显著差异,与DAVID L·MARKKS报导的密封水解24小时的效果相同。
A new method for measuring total protein in forage by acid hydrolyzed with ninhydrin is described. Standard Protein and forage samples were hydrolyzed with 3% sulphuric acid at electric stove immediately for 6h. Total protein were de Lermined by absorbance from hydrolysis with ninhydrin and pure protein were analyzed by differance of absorbance from hydrolysis and unhydrolysis all samples methanol extracts with ninhydrin. There were no significant difference in the results between semimicro-Kjeldahl elemental analysis and the method discribed. The results of this method were same as that Davids L. Marks's report.
